Axonis Therapeutics is chasing a harder and more elegant idea: restore the brain's own ability to inhibit itself. The company, founded in 2020 and headquartered in Boston, builds oral medicines around a single molecular target called KCC2, a potassium-chloride cotransporter that sits at the center of how neurons quiet down.

When KCC2 works properly, it keeps chloride balance in neurons tuned so that inhibitory signals land as intended. In 2020 they spun the work out, with contributions from Universite Laval, and Axonis was born.

How KCC2 keeps the brain in balance

Every thought depends on a balance between excitation and inhibition. KCC2 is the transporter that makes inhibition possible - and Axonis wants to strengthen it.

STEP 01

The set-up

KCC2 pumps chloride out of neurons, setting the electrical gradient that lets inhibitory (GABAergic) signals actually calm a cell down.
