Axel Hoos Keeps Choosing the Empty Blueprint

He helped give immuno-oncology a working language, rebuilt an oncology operation after its cupboard was cleared, and turned a young biotech into a clinical-stage company. Now Axel Hoos is starting again.

The recurring scene in Axel Hoos’s career begins the morning after the furniture has been removed. A scientific field lacks a common rulebook. A pharmaceutical company has sold most of an oncology business. A young biotech owns a promising platform but still has to prove that it can make actual drugs. Hoos arrives when the blueprint is incomplete, the vocabulary is disputed and the organization must be built while the science is moving.

He calls himself a builder. The word can sound pleasantly vague in executive biographies, but his record gives it edges. It means assembling a development framework around a new kind of medicine. It means deciding which programs receive money and which do not. It means translating among the laboratory, clinic, boardroom and partner’s conference table. It also means recognizing when the building can stand without you.

The field before the furniture

Hoos trained for a life at several borders. He earned his medical degree at Heidelberg University and a doctorate in molecular oncology at the German Cancer Research Center, working in Harald zur Hausen’s laboratory. Surgery training followed in Munich, then work in surgery, molecular pathology and tumor immunology at Memorial Sloan Kettering in New York. Later, Harvard Business School’s Program for Leadership Development added the explicit grammar of organizations.

That range mattered when cancer immunotherapy was still struggling against tools designed for a different sort of treatment. At Bristol Myers Squibb, Hoos and his team worked on ipilimumab, later marketed as Yervoy, the first approved immune checkpoint inhibitor. The drug was unusual, and so was its behavior. Immune responses could unfold on a different clock from conventional treatments. Familiar assumptions about when to look and what counted as progress were not always adequate.

Hoos helped spearhead a new development paradigm for the field. In papers and through the Cancer Immunotherapy Consortium, he worked with scientists, companies and regulators on common methods. The contribution was partly scientific and partly civic: a young discipline needed rivals to agree on enough rules that useful evidence could accumulate.

The consortium offers a small map of how he operates. Hoos helped create its predecessor at the Sabin Vaccine Institute, then helped transition the group to the Cancer Research Institute, where it could extend its reach. In 2011, CRI named him co-director alongside James Allison. In 2019, the institute gave Hoos its Oliver R. Grace Award for distinguished service in advancing cancer research. The career achievement was not a lone flash of insight. It was building the connective tissue that let a field learn faster.

Rebuilding the cupboard

Hoos joined GSK in 2012. Three years later, the company’s transaction with Novartis transferred its marketed oncology portfolio away. What remained was early research, scientific ambition and the mildly terrifying freedom of a near-empty cupboard. Hoos rose to lead the therapeutic area and chair R&D governance, with responsibility for technical and funding decisions.

The rebuild stretched across immuno-oncology, cell and gene therapy, epigenetics, synthetic lethality and direct tumor targeting. It included internal programs and a network of transactions: Tesaro, Adaptimmune, Lyell, Immatics, IDEAYA, Surface Oncology, iTeos and others. By the end of his tenure, GSK again had an integrated oncology operation, a substantial pipeline and approved medicines.

There is a practical idea hidden in that list. Modern drug development is less a procession of solitary geniuses than a carefully wired system. A large company needs enough judgment to know what it should invent, what it should license and with whom it should share risk. Hoos’s work sat at that junction. Scientific taste determined which doors were worth opening; governance determined whether anyone could walk through them.

In 2021, he left. The move was revealing because GSK was no longer the empty room he had entered. Hoos said he considered the rebuilding assignment complete and wanted to construct something new in biotech. Comfort, in this telling, was not a reward. It was evidence that the particular problem that attracted him had changed.

A toolbox, not a hammer

Scorpion Therapeutics was barely a year old when Hoos became CEO. Its premise, “Precision Oncology 2.0,” was an attempt to improve the selectivity of small-molecule drugs and expand the set of cancer targets they could address. The platform combined target biology, genome-wide CRISPR, medicinal chemistry, chemical proteomics, computation and translational medicine.

Hoos liked a hardware-store metaphor. Drug discovery had too often taken one attractive tool and turned every target into the same nail. Scorpion wanted the whole toolbox, then the discipline to choose the right instrument for each biological problem. “We’re elevating the whole game of small molecule creation,” he said. The point was not novelty for its own sake. He described his philosophy as operationalizing innovation: apply new capability where it can produce meaningful near-term impact.

During his tenure, the company grew to about 120 people and advanced three programs targeting mutated PI3K alpha and two forms of EGFR mutation. It formed business partnerships with AstraZeneca and Pierre Fabre and moved from early discovery into clinical development. Hoos departed the CEO role in early 2024. In 2025, Lilly announced an agreement to buy Scorpion and its STX-478 program for up to $2.5 billion, with the other assets and employees placed into a new independent company.

Urgency with a long memory

The Scorpion years show Hoos at startup speed. His Sabin tenure shows the complementary rhythm. He joined the vaccine institute’s board in 2006 and served for two decades, including six years as chair. When Sabin marked his departure in 2026, it credited him with helping clarify the mission, shift the organization toward a more entrepreneurial ethos and renew its research strategy.

Boards deal in patient architecture: leadership succession, mission, capital and the design of decisions that may not pay off for years. Hoos’s nonprofit record suggests that his fondness for building is not merely a taste for acceleration. He also stays. The common feature is an institution whose shape still matters.

His network reflects that instinct. He co-founded and served on the board of Australian biotech Imugene. He served on the boards of TCR2 and NextPoint Therapeutics, and helped found the PACT initiative associated with the Biden Cancer Moonshot. In 2025, Adagene appointed him executive advisor. Each position sits at a slightly different seam between scientific judgment, company design and collaboration.

A builder’s real product is not the room. It is the ability of other people to keep working after the scaffolding comes down.

Another launch

Hoos’s newest organization is Argonaut Bio, where he is founder, chairman and CEO. Public descriptions call it an oncology holding company developing a portfolio of next-generation medicines. The details remain spare, but the form is notable. A holding company is an organizational bet before it is a molecule: a way to gather assets, teams and capital under a repeatable operating logic.

The name Argonaut carries a whiff of adventure, but Hoos’s version of exploration has always been methodical. Pick the scientific territory. Establish the rules of travel. Assemble complementary tools. Make partnerships before pride becomes expensive. Measure what the biology is actually doing. The romance lies in the destination; the craft lies in packing the boat.

His career is easy to reduce to a chain of medicines and companies. More useful is the pattern between them. At BMS, an emerging class of therapies needed a development language. At GSK, surviving research needed an oncology organization. At Scorpion, a platform needed programs, partners and a clinical path. At Sabin, an enduring mission needed sharper machinery. Again and again, Hoos has occupied the unglamorous middle where insight becomes coordinated work.

There is also a lesson in the exits. Hoos left GSK after the oncology rebuild had produced a functioning enterprise. He left Scorpion before the later Lilly transaction supplied a neat financial ending to the company’s story. Neither transition fits the familiar founder mythology of clinging to the captain’s chair through every season. His choices suggest a narrower appetite: the stage when scientific possibility must become an operating system. Once the system has momentum, another unfinished institution can become more interesting than the one already running.

That middle is where biotechnology either becomes useful or remains an excellent presentation. It demands comfort with uncertainty but impatience with vagueness. It rewards people who can speak both mechanism and budget, both trial design and team design. Hoos has spent his career making those dialects converse.

Now there is another empty blueprint on the table. If the past is any guide, he will begin by asking which parts truly need invention, which can be borrowed, and which people must be in the room. Then the furniture will start arriving.
